Mechanisms of Protein-Protein Binding: Double-Mutant Cycle Analysis Using the ProteOn XPR36 System

چکیده
interactions. Schreiber and colleagues are using the multiplemutant cycle method to evaluate cooperativity of residue binding (Reichmann et al. 2005). With this method, the binding of two mutant proteins is measured individually and together; the interaction energy between the two proteins can then be determined by subtracting the free energy difference in the binding of a double-mutant protein from that of the two singlemutant proteins. The data presented in this tech note is in the context of this larger study.

متن کاملApplications of the ProteOn GLH Sensor Chip: Interactions Between Proteins and Small Molecules
Of the available ProteOn sensor chips, the GLH chip offers the highest ligand binding capacity, making it optimal for the study of protein-small molecule interactions. This higher capacity is attained through the structure of its surface binding layer, comprising a unique formula of modified polysaccharides. Higher binding capacity, together with efficient preservation of the protein’s biologic...

Here we provide data from a head-to-head comparison study using four biosensor platforms: GE Healthcare׳s Biacore T100, Bio-Rad׳s ProteOn XPR36, ForteBio׳s Octet RED384, and Wasatch Microfluidics׳s IBIS MX96. We used these instruments to analyze the binding interactions of a panel of ten high-affinity monoclonal antibodies with their antigen, human proprotein convertase subtilisin kexin type 9 ...
متن کاملMutant Profilin1 Aggregation in Amyotrophic Lateral Sclerosis: An in Vivo Biochemical Analysis
Introduction: Profilin1 (PFN1) is a ubiquitously expressed protein known for its function as a regulator of actin polymerization and dynamics. A recent discovery linked mutant PFN1 to Amyotrophic Lateral Sclerosis (ALS), which is a fatal and progressive motor neuron disease. We have also demonstrated that Gly118Val mutation in PFN1 is a cause of ALS, and the formation of aggregates containing m...
